Genome editing
Precise CRISPR-Cas9 editing of human iPSCs – from single nucleotide changes to knock-ins and knockouts, homozygous or heterozygous.
Consultation and project workflow
Every project can start with a free consultation. In this session we discuss your desired outcome and outline the approach we will use to achieve it.
Throughout the project we send intermediate reports to keep you informed of progress, discuss preliminary results and address potential challenges.
Genome editing tools
We use CRISPR-Cas9 in its ribonucleoprotein (RNP) form because of its transient presence inside cells. This offers two key advantages:
- Lower off-target effects
- Higher editing efficiency
Types of genetic edits
01
Single nucleotide changes
- We can introduce or correct single nucleotide variants (SNVs) of any type.
- Small insertions and deletions (indels) can be engineered homozygously or heterozygously.
02
Knock-ins
- Inducible and constitutive expression of Cas9 or transcription factors at a safe harbour locus (e.g. AAVS1).
- Reporter constructs integrated into endogenous genes at their native loci.
- Exon or cDNA sequence insertions into introns to rescue mutated genes.
03
Knockouts and knockdowns
- Achieved through frameshift mutations or targeted deletions of specific gene regions.
- Introduced homozygously or heterozygously, resulting in complete knockouts or partial knockdowns.
Quality control
Quality control of genome-edited iPSC lines is available on request. We follow stringent quality control measures accepted by the scientific community, including but not limited to:
- Genomic integrity evaluation
- Mycoplasma sterility testing
- Trilineage differentiation potential assessment
- Pluripotency verification
- Off-target analysis
- Deep sequencing of the on-target site
These measures ensure the reliability and quality of the edited iPSC lines.
